چکیده
Abstract Hair follicles (HFs) produce hair to mediate the first line of defense in mammals. HFs undergo cyclical production (anagen), and while this process is unsynchronized during adulthood, external cues, including shaving, induce synchronized anagen bolster surface barrier. Herein, we show that macrophages were crucial promoting anagen. Shaving triggered accumulation secondary germs (SHG), which underwent proliferation as early Day3 post-induction. Constitutive ablation markedly delayed anagen, targeted at Day 3, but not 0 or 6 post-shaving, impaired SHG activation with Anagen entry was dependent on Folr2 +tissue macrophages, CCR2 +monocyte-derived macrophages. Single-cell RNA-sequencing cell-cell communication analyses revealed +macrophages communicated SHGs via Igf1 (encoding insulin-like growth factor 1 IGF1). Ablation deletion IGF1 from decreased AKT signaling led both rescued injection recombinant vivo. ATAC-sequencing lack IGF1-from resulted alteration transcription motif enrichment chromatin accessibility various genes SHGs, demonstrating primed enter by IGF1R signaling. These results highlight importance immune-tissue crosstalk facilitating provide impetus for strategies promote growth.

background: the opening and closing of the implantation window is important for successful pregnancy in eutherians. the recent study demonstrated that the window of uterine receptivity was prepared by the sole action of progesterone in mice, but the mechanism to close the window remained to be elucidated. methods: the pregnant mice were ovariectomized on the evening on the third day of pregnanc...
متن کاملInduction of Hair Growth by Insulin-Like Growth Factor-1 in 1,763 MHz Radiofrequency-Irradiated Hair Follicle Cells
Radiofrequency (RF) radiation does not transfer high energy to break the covalent bonds of macromolecules, but these low energy stimuli might be sufficient to induce molecular responses in a specific manner. We monitored the effect of 1,763 MHz RF radiation on cultured human dermal papilla cells (hDPCs) by evaluating changes in the expression of cytokines related to hair growth. The expression ...

متن کاملInsulin-like growth factor 1 (IGF-1): a growth hormone.
AIM To contribute to the debate about whether growth hormone (GH) and insulin-like growth factor 1 (IGF-1) act independently on the growth process. METHODS To describe growth in human and animal models of isolated IGF-1 deficiency (IGHD), such as in Laron syndrome (LS; primary IGF-1 deficiency and GH resistance) and IGF-1 gene or GH receptor gene knockout (KO) mice. RESULTS Since the descri...
متن کاملInsulin-like growth factor I is essential for postnatal growth in response to growth hormone.
Insulin-like growth factor I (IGF-I) is essential for cell growth and intrauterine development while both IGF-I and GH are required for postnatal growth. To explore the possibility of direct GH action on body growth, independent of IGF-I production, we have studied the effects of GH in an IGF-I-deficient mouse line created by the Cre/loxP system. The IGF-I null mice are born with 35% growth ret...
